To help you understand the requirements to detect the desired minor allele frequency (MAF), we have developed the calculators below for use when planning your ThruPLEX® Tag-seq experiments. Remember that when determining the detectable MAF, there are several factors to consider including input amount, depth of sequencing, capture panel size and UMT family size. We advise using a UMT family size of around 8-10 raw reads to reduce false positives (1,2,3). For further explanation, please refer to our blog post about depth of sequencing.
